Nobody's mentioned webcomics yet, so I'm gonna list some of my favourites and give you general synopses. I want to know all your favourites as well! SPILL.
Penny Arcade - penny-arcade.com
Penny Arcade obviously doesn't need an introduction. It's just the ultimate gaming comic. Tycho and Gabe, comic representations of writer Jerry Holkins and artist Mike Krahulik, build three strips a week out of our favourite pastime. That appears to be the only rule. Also, there's a podcast (simply called "Downloadable Content") which updates, apparently, whenever they feel like it. It's always worth a listen if you're a fan, and for me it's definitely made me get another forty minutes or so of joy out of each strip.
Achewood - achewood.com
Chris Onstad's commitment to his comic and the world he's built around it is nothing short of OCD. Not only does he juggle, what, ten or so characters in th handful of strips he does in a week, he writes sepereate blogs on all their behalv..es? Is that a word? Can you pluralise it? Anyway. Achewood is my favourite webcomic. It's gotten to the point where I just understand it on a level so great that I can get a laugh out of almost every panel. It also won Time Magazine's award for the best graphic novel of 2007. That's ridiculous!
Dinosaur Comics - qwantz.com
Dinosaur comics is the finest example I've seen of minimum input, makimum output. Five days a week, Ryan North takes the exact same six panels of some MS Paint dinosaurs and adds some fresh dialogue. That's it. And it's hilarious. I can't believe he's kept it up for this long - there's well over a thousand now. The man is clearly a genius. You have to see it.
And now for the bitter hatred corner...
Ctrl-Alt-Del - cad-comic.com
I can't express how much I can't stand CAD. But if you read the appropriate post on a blog called "Your Webcomic is Bad and you Should Feel Bad" (Google it) or read Scott Ramsoomair's recent April Fools strip on vgcats.com, you'll get a decent idea of how I feel. Tim Buckley's success really makes me feel sick.

`I had no idea CAD coppied penny arcade! I've been a fan of CAD for so long... maybe it's because I never read Penny Arcade... I feel... betrayed a little =.=. I remember the first one I ever read was megatokyo... although I'm not much of a fan.
Have you guys tried PHD, Cyanide and Happiness, and XKCD?

`Cyanide and happiness is a riot! I'm a big fan of Toothpaste for Dinner, and Garfield Minus Garfield too.
As for long-form comics:
Here There Be Robots is really great fun

I loved Josh and Imp

`I read tons of these, love questionable content, but that was already said. So here are my three.
Three Panel Soul. Really great, random geek stuff from the guys who did MacHall. (removed)
Looking for Group. Five adventures go out to bring justice to the world .... sort of, I love the fact that like 3 out of the five have no problem killing. The elf however wants to do the noble thing all the time, but never works out. Then there is Richard the warlock, I love him.

One innovative one is MS Paint Adventure, The comics are set in a game universe. And you can suggest what any charactors does like a text based adventure game. I got hooked on Pickled Investigator and now they are on Homestruck
